This scientific poster provides an overview of the bispecific antibody discovery process using four HER-2/NKG2D bispecific antibody formats as a proof-of-concept case study. HER2 is a cell surface receptor overexpressed in certain cancers, while NKG2D plays a role in immune surveillance against tumor cells. The dual binding enables the bispecific antibody to bridge the tumor cells with immune cells, enhancing immune-mediated killing of cancer cells.
Multiple bispecific valences and geometries were sampled to ensure generation of a lead candidate with enhanced functional activity, as well as dual binding. The bispecifics were tested for efficacy, function, and safety in an end-to-end bispecific antibody discovery platform. Studies included target binding, off-target screening, effector function characterization, in vitro cytotoxicity, and in vivo efficacy models.
